ABSTRACT

Objective:To compare the immune responses to simply mixed and fused recombinant DNA vaccines of herpes simplex virus type 2 glycoprotein D (HSV-2 gD) and molecular adjuvant CCL19 in mice and to evaluate the protective effects.Methods:Gene recombination technology was used to construct recombinant DNA vaccines expressing HSV-2 gD and CCL19 alone or fused together. After verification by sequencing, Western blot and ELISA, BALB/c mice were immunized twice by intramuscular injection. Serum samples and vaginal lavage fluids were collected regularly after immunization. Splenocytes, mesenteric lymph node cells and rectal tissues were collected after immunization. Differences in humoral and cellular immune responses to the two forms of vaccines and their protective effects in mice were analyzed using end-point ELISA, in vitro neutralization assay, immunohistochemical staining, chemotaxis assay, vaginal virus challenge, fluorescence quantitative PCR, weighing and disease severity assessment. Results:The fused recombinant pgD-IZ-CCL19 plasmid could express gD protein and CCL19 protein in vitro, but the level of expressed CCL19 protein by pCCL19-IZ-gD plasmid was less than that by pgD-IZ-CCL19. The mice immunized with pgD-IZ-CCL19 showed higher levels of IgG in sera and IgA in vaginal lavage fluids ( P<0.01) and stronger neutralization ability than the mice vaccinated with pgD+ pCCL19. Compared with other groups, more lymphocytes were recruited in the rectal mucosa, the spleen and mesenteric lymph nodes of mice immunized with pgD-IZ-CCL19. Weight loss or disease symptoms were not observed in the pgD-IZ-CCL19 group after virus challenge. In addition, the positive rate of HSV-2 in vaginal mucosa and the mortality rate in the pgD-IZ-CCL19 group were the lowest. However, pCCL19-IZ-gD turned out ineffective in preventing HSV-2 infection. Conclusions:The fused recombinant DNA vaccine pgD-IZ-CCL19 could induce stronger immune responses in mice and provide better protective effects, which was superior to the simply mixed DNA vaccine.

ABSTRACT

Objective:To investigate the relationships between the expression levels of chemokines CCL19 and CCL21 in serum of patients with RA and interstitial lung disease,as well as the correlation between the chemokines and the clinical parameters.Methods:Patients hospitalized in the First Hospital of China Medical University from May 2016 to March 2017 were enrolled.Serum levels of CCL21 and CCL19 in the 62 patients with RA,among them,34 cases without pulmonary interstitial lesions while 28 cases with lung interstitial diseases,18 cases ⅡP and 20 health control were detected with enzyme-linked immunosorbent assay (ELISA).One-way ANOVA,LSD-t test,Kruskal-Wallis test,Pearson and Spearman correlation analysis were used for statistical analysis.Results:The levels of CCL21 and CCL19 were both higher in the RA patients than the health controls(P<0.05;P<0.01),The concentrations of CCL21 in serum of the RA patients with ILD were higher than thoes without ILD,higher than the health controls (P<0.05;P<0.01),serum levels of CCL21 in the ⅡP patients were higher than the health controls (P<0.01),too.The serum levels of CCL21 in men were higher than women(P<0.05).Serum CCL21 levels were negatively correlated with albumin and BMI(r=-0.280,P<0.05;r =-0.605,P<0.05).Levels of CCL19 were negatively correlated with the levels of C4 (r =-0.326,P<0.05),and positively correlated with the levels of D-Dimer(r =0.592,P<0.05).Conclusion:Chemokines CCL21 and CCL19 are highly expressed in serum of patients with RA,they may be involved in the pathogenesis of RA.Furthermore,CCL21 may has a correlation with the lung interstitial lesion in RA.

ABSTRACT

The chemokine CCL19 and its receptor CCR7 are widely expressed in the body by several types of cells,including neutrophils,macrophages and glial cells.The CCL19/CCR7 signaling pathway participates in the process of inflammation,homing of lymphocytes as well as the physiological function.Recently,more and more attention has been focused on the role of CCL19/CCR7 in the nervous system and related diseases such as multiple sclerosis and cerebral ischemia.However,the effect of CCL19/CCR7 on the central nervous system(CNS)is not clear.This paper reviews the relations of CCL19/CCR7 to the CNS,in hope of sheding light on the significance of CCL19/CCR7 for the nervous system.

ABSTRACT

Objective:To analyze the level diversifies of plasma CCL19 and CCL21 in the male drug users infected HCV.Methods: The plasma CCL19 and CCL21,anti-HCV and HCV RNA were detected by ELISA quantitation,ELISA qualitation and Real-time RT-PCR respectively.Compared with 60 healthy man,the level diversifies of plasma CCL19 and CCL21 in 391 male drug users conducted as part of HCV Surveillance Programme in Zhuhai were analyzed.Results: 180 of 391 male drug users were infected HCV and the infection rate was 46.04%.The level of plasma CCL19 and CCL21 in the male drug users[anti-HCV(-)/HCV RNA(-)] were higher than that in the healthy man (P≤0.001).The level of plasma CCL19 and CCL21 in anti-HCV(-)/HCV RNA(+) group was lower than that in the others(P≤0.05) .Compared with that in the healthy man,the level of plasma CCL19 and CCL21 in the drug abuse anti-HCV(+)/HCV RNA(-) group had significant deviation(P<0.05).Conclusion: Drug abuse can heighten the level of plasma CCL19 and CCL21 in man.Increasing the level of plasma CCL19 and CCL21 may conduce to spontaneous HCV clearance.It may prognosticate that HCV infection will be persistent and have a bad consequence when the level of plasma CCL19 and CCL21 were degraded.

ABSTRACT

Objective:To investigate the serum level of C-C chemokine ligand 19 (CCL19)and its clinical significance in rheumatoid arthritis.Methods:The serum CCL19 levels in both rheumatoid ar-thritis (RA)patients and health controls were detected by ELISA.The proportion of peripheral blood B cells and memory B cell subsets were also detected in some patients.Then the clinical and laboratory data of the patients were collected.The CCL19 levels in patients with different clinical features were analyzed. And the correlation between the clinical data,laboratory parameters,B cell subsets proportion and serum CCL19 levels were also analyzed.Independent samples t test,paired t test,Pearson and Spearman corre-lation were used for statistical analysis.Results:The levels of CCL19 was higher in the RA patients than the health controls (P 0.05).The levels of CCL19 were higher in the serum positive (RF and anti-CCP antibody)patients,but there were no differences between low and high disease activity RA,as well as early and non-early RA. There was no correlation between the serum CCL19 levels and the proportion of B cells as well as memory B subsets.All the proportion of peripheral blood CD27 + memory B cell subsets in RA was lower than the healthy controls,including CD27 +IgD +,CD27 +IgD - and CD27 + B cells.Conclusion:The increased serum CCL19 levels in RA patients are associated with the activity of B cells,so CCL19 might predict whether the RA type is a B cell mediated RA,and specify the treatment directions for the rheumatologist.

ABSTRACT

To observe the expression of CCL19 in colorectal carcinoma tissues and to investigate its effect on proliferation, migration and invasion of colorectal cancer cells. Methods: The expression of CCL19 in 85 confirmed colorectal carcinoma tissues and the corresponding adjacent normal tissues was detected by quantitative real-time PCR (qRT-PCR) and immunohistochemistry of tissue microarray. SW620 cell line highly expressing CCR7(CCL19 receptor) as screened by qRT-PCR and Western blotting analysis was stimulated by different concentrations of rh-CCL19 (0, 10, and 100 ng/mL). Then the cell proliferation, migration and invasion capacity were examined by CCK-8, wound healing assay, and Transwell assay, respectively. Results: Immunohistochemistry and qRT-PCR results demonstrated that CCL19 expression was significantly lower in the colorectal carcinoma tissues than in the normal tissues (P<0.05). CCL19 expression was associated with tumor size and invasion depth (P<0.01). Treatment with rh-CCL19 significantly decreased the proliferation, migration and invasion capacity of SW620 cells(P<0.05). Conclusion: CCL19 expression in colorectal cancer tissues is lower than that in the adjacent normal tissues, and the expression is associated with tumor size and invasion depth. CCL19 can inhibit the proliferation, migration and invasion capacity of SW620 cells.
